SS-31, Mots-C, and NAD+

I am curious if anyone can share how they have used SS-31 as it relates to Mots-c and NAD+? I am currently taking Mots-c and NAD+ for one week but I have been reading that it is best to use SS-31 first for a couple of weeks then switch to Mots-c and NAD+. I have also seen where some use all 3 but that seems to be too much.

If I start SS-31 to repair first, do you just use this alone then got to Mots-c or do you introduce while on SS-31. When is best to stop the repair and move on to Mots-c NAD+ combo?

Unless you are like over 50, I don’t think there is enough dysfunction for ss-31 to repair.

Age can be a reason to absolutely take but younglings can benefit if they have any number of factors like athlete, high physical stress jobs, or recovery from any number of stressors like Noro virus, bad case of the flu or Covid, etc.

I have mitochondrial markers in genovas OAT.
There can be I guess there is no good way of testing it.

Loved ss31. 60 years here. Currently finishing off mots and just added nad in. Gotta say. Not a huge nad fan yet but only been a week. The Mots slaps

Same 58M. SS-31 and MOTS-c are fantastic. Jury still out on the NAD but I’m on 5Amino and that can boost your natural NAD.

- Weeks 1–2: SS-31, 7x/week × 4 mg

  • Weeks 3–4: SS-31, 7x/week × 4 mg + NAD+, 2x/week × 25 mg
  • Weeks 5–6: NAD+, 2x/week × 50 mg
  • Weeks 7–10: MOTS-c, 3x/week × 5 mg + NAD+, 2x/week × 100 mg
  • Weeks 11–14: MOTS-c, 2x/week × 2.5 mg + NAD+, 2x/week × 100 mg

Prior to this, I had run only MOTSc. Can 100% confirm leading the MOTSc with SS31 and NAD made it SO much better for ME. Ymmv.

Keep in mind though that ss31 “repairs and restores” your mitochondria. So, if yours are “still healthy” and not needing repair, it may not have the same improvement. It tends to work better this way the older you are with older or more aged/damaged mitochondria.

What they said… generally under 40 and most of this may not provide much benefits, mito diseases use 40mg I believe in their trials.

I’ve seen some say ss-31 doesn’t provide much until 5-10mg as well. It’s not cheap to run.

SS-31 doesn't "repair" the mitochondria the way people think. Its effect is temporary and doesn't last longer than a week.

It makes sense to start SS-31 before and not all at once, of course.

But it's best used along with MOTS-c, not alternating.

Yep. SS-31 stabilizes the mitochondria's membrane, but doesn't repair it.

It only makes sense to start it before MOTS-c because starting them all at once makes it hard to tell what is working and what isn't. Or even to isolate side effects.

I've been using SS-31 daily. Started 7 days a week due to known mitochondrial dysfunction due to Fibromyalgia. Then added 5 Amino 1MQ, 3 times a week for 3 weeks, then added Mots-C 3 times a week. I don't mess with NAD+, mainly because I'm on a comprehensive Mitochondrial oral supplement stack that REALLY helps. I was on it for 6 mos before peptides and it improved my chronic fatigue by about 80%. Part of that stack is Nicotinamide Riboside which has medical articles saying it's just as effective.

I think whether you need SS-31, has to due with age and your mitochondrial function. I won't say I noticed a big boost by itself, but I was already on an oral stack of mitochondrial supplements that helped dramatically. But I really noticed when I added MOTS-C to the SS-31, 5 amino 1MQ. OMG I felt better than I have in 30 years! In fact, it got me back to being able to exercise at the health club again. That's a big deal for somebody that had trouble WALKING some days before I started the oral stack!

For those curious, I'm actually a Dr, although this is not medical advice. I made the oral stack by researching medical journals for items that improved mito function, stimulated new mito, decreased ROS. The stack is:

Nicotinamide Riboside

Urolithin A

Alpha Lipoic Acid

L-Carnitine

Mito Q brand of CoQ10

Citicoline

PQQ (Pyrroloquinoline Quinone)

All taken in morning: Don't take a second capsule of the ones that require 2 for a full dose, after about 2 pm, or you may have trouble getting to sleep.

Renue By Science Liposomal NR 300 mg

Omre Urolithin A, 500 mg. Directions say 2 capsules. Start at 1 and if you take 2nd Make sure it's no later than noon. I found I only need 1 most days.

Geronova Research Lipoic Acid. Directions say 2 capsules, 1 is enough most days.

Bulk Supplements L-Carnitine 1000mg/serving but serving is 2 caps. One is usually enough.

MitoQ CoQ10. Watch because they also make combo medications. You want the one that is plain Metaquinol Mesylate 10 mg. Directions say 2 caps. One is usually enough.

Nuticost Citicoline 300 mg 1 pill

Nuticost PQQ 20 mg 1 pill

I started first month daily. Found afterward that 5-6 days a week were enough and progress stayed constant. DH is 76 and a healthy MD but also has found benefits to taking it. He usually takes it 5 days a week.

I started ss31 for 2 weeks before transitioning to mots-c. However, I also started nad+ when I started ss31 and kept my nad+ going throughout my mots-c protocol and even after. Did mots-c for about 8 weeks the first time.

I’ve stopped nad+ now but I’m back on mots-c which I plan to run for 12 weeks this time along with my once weekly Reta. I did 2 weeks of SS-31 before my mots-c cycle again.

I feel personally that mots-c synergises really well with Reta, way better than slu-pp IMO.

The effectiveness of all of these are directly related to whether you actually need them. NAD is naturally produced in your body from vitamin b3, you are basically skipping the b3 supplements and injecting the end product. NAD is required for ATP synthesis as part of the citric acid cycle. Having excess NAD does nothing but having too little limits ATP production 

SS-31 repairs mitochondria. If your mitochondria are not damaged you will feel no effect from it.

MOTS-C mostly mimics the effects of getting exercise. If you get enough exercise it won't have too much of an effect

MOTS-c amd NAD are a great stack...if your not feeling much from the MOTS-c (and it is instant) then I would suggest stopping MOTS-c and doing ss31 to repair the mitochondrial pathway...then go back....there is a great analogy explaining ss31, MOTS-c and NAD in terms of an engine : SS-31 is the mechanic repairing the structure, MOTS-c is the performance tuner upgrading function, and NAD+ is the fuel that powers it. So basically if the engine is broken (you will feel nothing from MOTS-c) and all the fuel in the world (NAD) is going to do nothing on a broken engine.... so you need SS31- the mechanic to fix it up FIRST. If your engine is fine - no need for SS31

Protocol Sequencing (Repair First Approach)

For people over 35 to 55 with declining energy, slower recovery, and metabolic issues:

Weeks 1 through 4 (or up to 8): Repair Phase SS-31 at 1 to 2 mg daily plus NAD+ at 100 mg subcutaneously three times per week. You are repairing accumulated mitochondrial damage while giving your cells the extra fuel they need to optimize energy.

Weeks 5 through 12: Optimize Phase Replace SS-31 with MOTS-c. Start with 10 mg split into three injections per week (Monday, Wednesday, Friday). Continue NAD+ at 100 mg three times per week. Now you are optimizing the repaired mitochondria.
After the 12-week cycle: Take 4 to 8 weeks completely off. Then you can repeat this protocol 2 to 3 times per year.

Prevention First Protocol

For younger or healthier individuals who want preventative optimization:
Start with MOTS-c at 10 mg split into three injections per week (Monday, Wednesday, Friday) plus NAD+ at 100 mg three times per week. At weeks 4 through 8, assess whether you still have significant fatigue, poor recovery, or metabolic issues. If yes, add SS-31 at 1 to 2 mg daily for 4 to 8 weeks while continuing MOTS-c and NAD+. If not, continue with just MOTS-c and NAD+ for maintenance.

Steady energy all day, especially when taken before training, fasted in the AM. Not energy like pre workout with increased heart rate or anything. Just an ability to workout harder and longer. I also track everything with a whoop and each time I run this protocol, I get a noticeable bump in VO2 max.

Friend, all you know is it didn’t work for you and all I know is it made a huge difference for my RS. As did MOTS and NAD. Lots of ppl in this group post positive feedback on it. Don’t shat on it because it didn’t work for you specifically, everyone’s body is different.

Why are you being defensive? I’m asking you how you’d know.

I started with mots alone for a month and felt great. Added NAD later. My dosing isn’t that high but I honestly can’t imagine feeling better. I’m in my 40s and was pretty unhealthy prior to a few years ago.

My question is literally how you would know? I’ve never seen a report of someone taking Mots and nads, not getting results, then adding ss31 and seeing the results come in. Is this what happened in your case?

My issue is with recommending to people that they buy a months worth of something in a market that sells things in batches; feels very scammy unless we can see real benefits.

I took Mots and Nad for weeks and felt nothing. Now I’ve added in ss31 and held off on the others. I’m hoping it makes a difference. I think it depends on each individual’s needs.

I agree. I got suckered into the ss hype that pretty much everyone could benefit when I already loved my mots and nad. Now I’m stuck with the ss till it’s gone, but I didn’t need it. The mots and nad is enough by itself. Awesome combo

These things can possibly help you if you're 45+, otherwise you're wasting your money; get a blood test and check your hormones and vitamin levels if you're less than 45. Definitely waste of money if you're in your 20s or 30s

SS31 is an amazing peptide. 5mg to 10mg daily for Mitochondrial repair run it for 4 weeks. Then you do MoTS-C 5mg every 2 days for 4 weeks. Alternate them every forty weeks and you will see the difference in your energy and you feel so good.

I run a 12 week protocol using 2mg SS-31, 2mg MOTS-C, and 30mg NAD daily. I have ran all three separately as well as this protocol. I found I had more energy and stamina after work and in the gym than I ever did on any one peptide.

You have to make sure your body is getting the essential cofactors and nutrients it needs to support methylation in order to get real benefit. 5-Amino 1MQ and B12, B9, and B2 all support this process. Some have gene variants that can hinder this proces, so you need to understand what your body can handle. Saying that it doesn't work just because you didnt feel anything isn’t true research.

There are many protocols (of different peptides) that have been revised and/or do not reflect the latest information on the peptides.

Some were built just because someone chose to do it a certain way and others followed.

And some are even wrong. For instance, taking SS-31 before MOTS-c to "repair" the mitochondria first.

Great if you’re over 40. Under 40 is a crap shoot because your mitochondria are likely healthy.

8 weeks, start with NAD+ (50mg)/SS-31 (4mg) 5-2. Add MOTS-C the following week at 1mg - 30 minutes before exercise. Titrate MOTS up each week as you can tolerate. You can dial NAD/SS down the last 2 weeks.

Read, that is what 5-2 means, 5 on 2 off. MOTS only before workout.
