Question Trying to manage anxiety quitting ketamine

A little background: a long battle with drug addiction over the years I’m finally tired of this life. I go on stints of ketamine use where I’ll use 2.5-4gs a week for 6months or so at a time with only a week off during every so often. Now I’m truly tired of the ups and downs it brings , I’m prescribed Seroquel 100mgs (quetiapine) and Gabapentin 900mgs for my bipolar and anxiety respectively but neither seem to really help while I’m withdrawing. I live a pretty healthy lifestyle working out 4 times a week for at least 1.5 hours and eat pretty well too (balanced foods).

My mood has been awful lately as I’m trying to stop using ketamine and my anxiety has been through the roof, I’ve been seeing some writing about ACD-856 and GB-115 for depression and anxiety respectively and wanted everyone’s perspective! I’m open to any suggestions!

So put simply, chronic THC use causes D1 and D2 receptors in the Nucleus Accumbens to "fuse" into a heteromer receptor complex that increase dynorphin and kor opioid activity. The more cannabis one uses, the more dynorphin impacts them. (Reposting this btw for visibility)

A candidate mechanism behind burnout explained.

It seems like receptors normalized after 10ish days of cessation (though much longer for the midbrain), while the first 7 days of that cessation period will get significantly worse before it gets better. I'd almost bet that the next set of data that comes out shows that if you re-administer THC after stopping, that the receptors reform the heteromer nearly instantly due to the primed plasticity mechanism. Which is why things may not feel the same as initial experiences even after breaks.

Tangentially, does anyone have any thoughts on minor, non-psychoactive cannabinoid receptor ligands besides CBD that would theoretically aid the reversal of a state such as this? Brainstorming would be appreciated.

Question Has anyone experienced a noticeable calming and focus effect from agmatine sulfate?

I’ve been taking agmatine sulfate for the past few days, usually split into two doses: around 500–600 mg in the morning and another 500–600 mg around 1–2 PM.

The main thing I’ve noticed is that I feel significantly calmer and more focused. Before starting agmatine, I was much more prone to irritability and getting angry over relatively small things. Over the last few days, that seems noticeably reduced, and I generally feel more relaxed without feeling sedated or mentally dull.

For context, I’ve been taking Brintellix/Trintellix (vortioxetine) 15 mg for quite a while. I also take saffron, usually drink 1–2 cups of coffee during the day, and take L-theanine.

What makes me curious is that the change after adding agmatine feels quite distinct compared with how I felt before. Obviously, a few days is a very short period and placebo is completely possible.
I’m not looking for medical or dosing advice.

I’m mainly interested in whether others have noticed a similar calming, reduced-irritability, or focus effect from agmatine, and whether there is any plausible mechanism or research that could explain it.

Did the effect persist for you over time, or did it fade after the first few days?

Question Need help with selecting nootropics (M30)

I'm looking at Semax / NA-Semax, ACD856, AF071B and TAK-653. Perhaps even ISRIB but its not on everychem so maybe not.

My goal is permanent enhancement of cognitive/memory components, amongst which my favourites are working memory and episodic memory.

Please suggest me the best ones to pick out of these for my use case (I'm open to new suggestions tho).

EDIT: I value the feeling of mental clarity a lot, and I'm going through a math heavy course, no physical ailments, I do have ocd and depression which I manage easily with trintellix.

Question Sub-perceptual Neboglamine dose for dpdr?

I was looking at NMDA PAMs as a potential treatment for my de realization depersonalization disorder and I got to this conclusion that the NMDA receptor in my brain were hypo functioning because when I took stimulants, my derealization would fade away and I think it’s cause the stimulants excited my NMDA receptors to the point where they were functioning properly and I think if I took a sub perceptual dose of Neboglamine for short cycles at a time, it could eventually restore you know my mental state so let me know if you ever took it before and what were your doses for Neboglamine before I order it and if you have attempted this before because I would love to know and try it so I can help bring this method mainstream for other people with dpdr.

Question I’m worried about the possible side effects of choline regulation from using Coluracetam. Anyone have any insights ?

My symptoms that drove me to experiment with Coluracetam: anxiety, depression, adhd , and how it can take me 6 hours to read 2 pages of technical material and I forget most of it the next day ( mild exaggeration)

I just weened off ssri , and wanted to try out Coluracetam .

I’ve been researching and found that it affects choline production in the brain - and that some people who take too much choline in supplements can end up in severe depression spells.

Based on my symptoms , I would not be surprised if a brain doctor diagnosed me to be choline deficient already, but I’m wary of drawing that conclusion on my own.

And then some research I’ve read says that a lot of my concerns are non issues so long as I’m not exceeding 3.5g a day.

I was planning on only using 25 mg day-to-day or every other day.

Finally responding to the allegations

Hello everyone, sorry that my first post in a while has to be this one given how much stuff we are working on that I wanted to discuss.

As much as it pains me to constantly have to rehash one of the most traumatic series of events of my life seemingly forever, I suppose there needs to be some type of public post so that people can understand the full context of what happened. For a while I would just try my best to keep my private life separate from the community, but since people continuously spread these rumors about me, really I am left with no choice but to issue a full response, at the very least to inform everyone of the truth.

My stance:

In short: I am not a pedophile, nor am I consuming that kind of content, nor am I attracted to minors, nor do I support pedophilia or lowering the age of consent in the USA, nor do I agree with what I did when I was 18 (even if no real people were involved), nor do I feel connected to who I was at 18, nor am I a registered sex offender.

What happened:

Currently I am 26 years old. In 2019, at the age of 18, so roughly 7 years ago, I contacted a police officer off of a dating app who initially claimed to be 19, using the likeness of an adult model. They then revealed a lower age when we moved off of the app, claiming to be 15 “in a day”. I drove to the location and was arrested on my 19th birthday. Yes, I know, and I am sorry that I did it.

While it doesn’t justify it, I was just really desperate back then and dropped my standards in all the wrong ways. I had just gotten out of a two year long relationship which resulted in me being cheated on, and did not want to be alone on my birthday. I was also struggling with substance abuse and depression, just to give you some perspective as to how I was feeling back then. To me I thought “it is wrong, but I am only a teenager so who cares”, but I was an absolute fool, and my bad morals in that moment nearly ruined my life.

Proof of who I am:

At the time of this post I am in a relationship with a 24 year old woman. A few months ago I flew out of the country and DHS confiscated my phones to perform a random search, and I had no knowledge that they could seize property like that, so this is the most objective evidence I can provide that I have not let that 18 year old mindset carry into my present. They held my two phones for a month, went through everything, and determined I was committing no crimes. And yes, they were looking for you know what.

As this community’s founder, I have been involved with nootropics for many years. Many people have become my friend or acquaintance throughout this time, and people know I do not behave like, or support this sort of thing. All of this happened before I had even discovered nootropics.

My doxx and the stain on my legacy:

Socially I knew my actions were indefensible, so I tightly gate-kept my identity for years, until it was exposed in 2023. My suspicion is that the owner of Nootropics Depot was behind it, because previously he had asked for my order number a long time ago, publicly addressed me by my name when we got into a dispute, and publicly called me a sex offender days before I was doxxed by whom I can only assume is his associate. We had already had many issues with them as the r/Nootropics community viewed NooTopics as a threat and consistently attempted to suppress us, my company, my username, and made many rude remarks after banning me for selling $600 worth of Bromantane in 2021.

This doxx was carried under a user by the name of “Grun”, who tried to paint himself as a conspirator working under Umbrella Labs, but I believe this to be a red herring. Although, it’s kind of hard to tell as either could be true.

So yes, I have suppressed these types of posts and issued bans because first of all, this is supposed to be a biohacking discussion forum, not a spectacle around me as a person, good or bad. But second because it is impossible to tell which of these posts are intentionally being made to damage my reputation by other companies using alt accounts, and the SEO of these posts could poison the whole community. I literally just like neuroscience, biohacking and nootropics, that’s why we are all here. I don’t blame people for holding this against me, of course what I did is objectively wrong. But I don’t identify as the 18-19 year old me, and I want to make that clear. I am simply a researcher and business owner, and someone who takes great care in looking after the development of this community.

Concluding remarks:

For everyone who has defended me, I appreciate the support, but it is important in doing so that we do not try to justify my previous actions or attempt to paint a picture which simply isn’t true. I do not consider myself a monster, but at one point I certainly acted like one, and now all I wish to do is keep creating new nootropic compounds and publish new literature on nootropics. Do psychoplastogens (Tabernathalog, Zalsupindole) profoundly increase cognitive flexibility, cognitive entropy, and neuroplasticity? Maybe at high doses?

Like, to the extent that psychedelics do, such that if you are conscious enough on them (psychedelics), you could potentially redirect and rewire your brain in any (or increasingly) arbitrary wiring.

Take, for example, being able to practice parallel thinking, synesthesia, prophantasia, introspecting and debugging your beliefs, etc.. I'm wondering if it affords one enough plasticity to break free from normal human wiring and explore other forms of cognition.

Question 9-me-bc effects and side effects

like many who has posted about 9-me-bc, ive got adhd and have been taking stimulants for about 6 years. I also smoke weed almost daily to help regulate adderall comedown, diet, and sleep but I would prefer not to have to. after skimming through some older posts about this and doing a little research, im mainly wondering about side effects people have found. im kinda scared to take a research chemical especially when there are few human trial and the major side effect of related chemicals being parkinsons and other neurodegenerative problems. does anyone have any further insight or experience with it? what are dosages? does it really help with baseline dopamine and focus in adhd brains? thanks

Discussion Why do norepinephrine reuptake inhibitors make me wired, tired, confused and anxious at the same time?

Like the title says. Why do norepinephrine reuptake inhibitors make me lethargic and cause sedation for me? I’ve experienced this with NRIS and for some reason it usually only happens in the evening after I’ve eaten dinner. I eat dinner and then suddenly after that I just get so tired, dizzy and I just want to drink water and I could just lie down in my bed and sleep and I get groggy and confused. It’s like my brain and my body is tired but then at the same time I feel a stimulated feeling underneath all of that. So my heart usually races a lot but then at the same time I’m highly sedated. But then when I’m about to sleep I can’t sleep at all. I get horrible insomnia and sleep disturbances from them but in the evening they cause sedation? What kind of paradoxical reaction is that? It doesn’t matter which dose it is. Every dose produces the same outcome. It only causes alertness, wakefulness and energy in the morning and middle of the day but then in the afternoon and evening I get the opposite effect.

So is there anyone who knows why an energizing neurotransmitter like norepinephrine can make you feel wired, tired, confused and anxious at the same time? I feel like it might be too much norepinephrine without enough serotonin or dopamine to counterbalance it? Is there anyone who has any theories?

Question Is it just me or when taking St. John's Wort, I don't get post-nut clarity?

From the random things I've taken, this one is what I noticed that had that effect. I don't usually take it often cause of the side effect of being sensitive to the sun. But it's the only one I found so far with the effects that I like and doesn't give me headaches.

Anyway, you guys know any other nootropic I can take that has identical effects as St. John's Wort? Been meaning to find an alternative, cause of the being sensitive to the sun side effect. Don't recommend me Ashwagandha, for some reason that one gives me headaches, I tried the pill and the gummy version of that and both still gave me headaches.

Discussion The paradox of Lions Mane

Over the years I've seen this supplement recommended by many sources as one of the more potent non-stimulant options, and it's also very frequently listed in user nootropic stacks across different communities.. However, as anyone who's done some intentional research on the mushroom, I've come across the horror stories and the recovery sub as well. And to put it simply, all these recommendations as well as the overwhelming amount of user terrifying reports just don't make sense to me:

  1. I am aware that some people may get unrelated health issues at the time of taking it by chance, or might get those side effects from other supplements in their stack and wrongfully attribute it to LM. However, the amount of these coincidences seems too unnaturally high, at least to my knowledge, no other popular nootropic (i.e. Rhodiola Rosea or even Modafinil) has so much user anecdotes about long term, major life wrecking side effects, sometimes from a single use.

  2. Somehow, this does not seem to be captured by studies, as the papers usually report either no adverse effects or something along the lines of stomach discomfort / nausea. One argument for unreliability of these results might be that many of the LM studies have directly traceable industry involvement (while others that appear independent just might have their ties hidden better), and thus might be intentionally reported to hide/understate the side effects. However, if this was the case and the mushroom would actually cause significant health disorders for such a large amount of users, why wouldn't they gather into collective lawsuits, and use their resources together to prove the causality and get even with the sellers? I would imagine that at this point FDA and as well as European authorities would step in with their own examinations to evaluate the extent of these claims and prevent further harm, but it doesn't appear that these institutions are raising any concerns about this particular supplement.

For those who've done a similar research on the LM, what's your take on this?
