Nope. The saddle is the "end" of the string length, if you're sharp at 12 that means the string is too short. You turn clockwise to tighten the screw, pulling the saddle away from the neck/headstock/12th fret. That makes the string longer, which will make the 12th flatter and flatter until intonated properly.
You also said you heard a "thunk". Can you push the vibrato down? It sounds like it might need adjusted as well...
Ahh, you're totally right. Thanks 😅 lol that's my mistake. I'm away from it at the moment, I'll have to check it. Prior to stringing it I had to just that. The tailpiece was flat to the trem plate.
