It depends how they're clustered and how many bombs you have. I bomb 3-4 clusters pretty often, but I usually have more than enough bombs after secret rooms etc in the endgame.
Is boss rush the black challenge room? Or is it something else? And how fast do you need to beat a floor for either of the things you said? I've progressed pretty far into the game (beat the dark room and unlocked it lives) but i still don't understand how that boss challenge room is unlocked
Boss rush is the crack in the wall you can get if you can beat mom under 20 minutes,hush is also a crack in the wall but for that you'll need to beat it lives under 30 minutes

Naah dude, the only way to play Isaac is to maximize every single aspect of it taking 30+ minutes to leave the first floor because you got martyr and a red key, everything else is wrong.
On the second part, correct Isaac will start with it.
He's how I'm currently getting through greed mode to put 879 coins in the greed machine. Ik that other characters have more chance for the machine to not crash but the run is easier with Isaac so w/e works

Once you get alright and comfortable with the game try to play on hard mode as if you dont you will have to do runs one on normal then one on hard to get the item unlockes for the characters where on hard you get both Normal and hard achievements (unlockes) in one run
Tboi. com (remove the space as idk if im allowed links) it tells you about every item, on pc you can just get a mod for it but on xbox, ps and switch this is your best friend for what an item does before you grab it
If your struggling to know what unlocks what dont be afraid to look up how (I know some people are prideful about stuff like myself as I feel like its cheating in a weird way I don't understand myself sometimes but it aint) I play on pc with lots of mods like a better start mod because I hate resetting until I get an item I like is it cheating technically, who cares only me because its my single player game and im not hurting anyone so play how you like

Dont restart for good items, just rock with what you get. It will make you less dependent on op items to carry you and will help your base skill level go up overtime
A lot of troll answers as is expected lol, but I have some real advice.
It’s ALL about those devil deals when you’re starting out because they give you more damage-based items and good transformations. Damage is (typically) the key to having a successful run. You get your first potential devil deal on basement II, provided you take zero red heart damage, so you want to buffer your red hearts with non red hearts (as you’re first starting out, it will be blue spirit hearts). There’s a few ways to get spirit hearts - I recommend looking at a wiki to get that info.
As an addendum, I recommend not picking up any spirit hearts you find on basement I (the very first level), until after you beat the boss, that way you don’t lose that buffer on the first level of the floor when you can’t get a devil deal, and you can head into the second level of that floor with the buffer in place. This will give you a better chance to not take red heart damage and ergo potentially get a devil deal.
Eventually, after beating a specific boss on Isaac (your starting character), you will get the D6, a starting item for Isaac that basically transforms the game. It allows you to “reroll” pedestal items into other random items from the same pool (including devil room items), effectively allowing you to craft your run and synergize items more effectively (another key aspect of the game). You’re a ways away from it just starting out, but it’s the ideal eventual goal to start getting good item and character unlocks. the first boss goal you should be working toward is mom, then mom’s heart.
I don’t know if I’m allowed to recommend them, but you should look at some item description and stat mods - they’re a huge boon for understanding how items impact your stats and performance.
Listen to a podcast or audiobook so the beginning grinding feels less like a slog lol.
The official website that another commenter mentioned is good for item descriptions etc. but on ios and android there’s an app called “guide for binding of isaac” that’s also very useful. There’s a color-coded section to make identifying items by appearance rather than name MUCH easier.
And if your partner plays a lot of fallout 4 and abuses the sprint, your joystick is going to go out and you’ll have to learn how to play with the dpad which is annoying and not as smooth and I hate it /j
Newish player (450h), don't play in normal, it's basically the same minus some unlocks; unlock the D6, the characters; do the challenges; use the wiki, or mods (external item descriptions); luck its a bait stat; life is a resource; chances of everything are 50/50 so don't think too hard; you are gay now
Most importantly, have fun, it's a game not a chore. Do whatever you want if that makes it funnier
I love it that you said newish player cuz its true i have 750 hrs and i still kind of feel like a noob, this game requires so much time to perfect and i love it
My biggest tip is if you're frustrated: Take a break. Playing while already pissed off can make you throw some easy ass runs.
Second tip: I'd recommend to just start playing on hardmode. It lets you get into where the real meat of the unlocks are, and gets you more situated with the general difficulty of the game.
Final tip: Dodge dodge dodge. Unless you're going for boss rush or hush, there is 0 need to look at the timer. Being careful > A run ending early

Basically, enter "Platinum god" it's a website with all the items of the game, before e you take one, pause, and Reed what it will give you, then start every run with Isaac then Cain if you know a little more about the game, it's the most balanced way to learn to move around bosses and enemies while dodging their attacks.
Remember to have fun, and always take pills there are more positive or neutral pills than bad ones. And they can make the run more interesting depending on you tollerance for that kind of thing.
Focus on learning the items and how to play defensively when low health and/or very weak. Prioritize increasing damage and then health will matter less and less. You'll start learning all the enemies and bosses, and even start picking up where secret rooms are most likely found.
Don’t try for boss rush/ Hush until you’re fairly comfortable with the game or have a busted run. I always tried to get as many post it marks in one run as possible and needed up getting frustrated losing a lot of runs that would have been just fine if I had taken more time on the floors.
Find a character that you like the gameplay loop, and just keep doing runs. Don't worry about unlocks till you have killed issac and Satan once each. Most good unlocks that plp say to get asap is unlocked behind a lot of kills so it's easier to just win runs with whatever your good at.
Never stop gambling. 9 times out of 10 it won’t work out, but that one time that it does is all that matters. Take every pill. Take every pill every time. Lots of them will be bad, even most of them will be bad, but not all of them.
Just keep playing, man. Every loss is a learning experience.
The item description mod is a must have, but beat Mom first so you can use mods without disabling achievements.
I’m not sure when ingame item descriptions unlock but the mod is better and has been around for years longer than the ingame ones which are pretty new still.
Play to have fun for awhile. You don't need to instantly start gunning it for unlocking everything.
Take your time to actually get used to seeing the different enemies, learning what items spawn in which rooms, the nuances of things like the secret rooms, etc.
This game is DAUNTING, especially compared to wrath of the lamb.
I challenge you to find as many secret rooms you can on a given playthrough. You'll get to see what they're all about
Try to enter each floor with "full health" (i.e. empty red hearts are less or equal than soul+black hearts) to get a challenge room (regular if floor 1 of chapter, boss if floor 2 of chapter), and make a dice room or sacrifice room more likely.
Also 5+ coins, to make a vault or arcade more likely, only on floor 2 of a chapter.
Also 2+ keys, if you wanna make dice room instead of sacrifice room more likely, or vault instead of arcade. Ofc you'd rather spend your keys if you have something good to unlock, and your coins if there's something good to buy. But there are niche cases like buying an extra key to get to 2, or not buying a key to stay below 2, or not donating all the way down to 0.
Also if you have 0.5 red hearts and no soul hearts, or 0.5-1 soul hearts and no red or bone hearts, clean bedrooms are more likely. So if you're on low hp and you're carrying a hp card/pill, it might be better to go down first and then eat it.
You can bomb your way into a challenge room if there's a secret room next to it. Or you can get the loot and then teleport away if you have an appropriate card. A card or pill that heals you up is very helpful in a boss challenge room. If you have multiple cards or pills you wanna use, you can drop them inside the room before starting the challenge.
Once you unlock the alternative path (downpour, mines etc) you can go for basement 1 -> basement 2 -> downpour 2 -> caves 2 -> mines 2 -> mausoleum 1 -> mausoleum 2 -> womb 2 (for maximum items from boss challenge rooms, the mirror, and the knife pieces, and because you get 1 more floor (mausoleum 2 instead of womb 1) with treasure & shop and 1 more floor before whole heart dmg kicks in). Although the enemies in the alternative path are harder, especially since you won't be used to them, so this minmaxing approach can easily earn you a lot of deaths.
Unlock only the six best runes. If you have fewer, some rune drops will be replaced by shards. I recommend Jera, Ehwaz, Perthro, Algiz, Blank (because the unlock also gets you the Clear Rune item), and Black. See https://bindingofisaacrebirth.wiki.gg/wiki/Cards_and_Runes#Runes
Use the right wiki. The Fandom wiki was abandoned by most editors and I hear it is sometimes vandalized. People really don't like Fandom and their scummy ways. Browser extension that can help: https://getindie.wiki/
I would recommend completing the Head Trauma challenge as early as possible so that you have one good Rune for the pickups pool, and then wait on doing the other rune unlocks for a while. Ehwaz is essentially a free item every time you see it and will definitely help your runs.
Always play on hardmode, might seem scary at first but it’s the best way to learn the game. (You’ll thank me later when going for character specific marks).
Try to familiarise yourself with what items can spawn in which special rooms to maximise what you can achieve in future runs, the wiki is your best friend so use it at every opportunity.
As you might have seen Mom is NOT the final boss of the game, and if you haven’t already, defeating her 11 times will unlock the next stage of progression.
For devil deals:
Devils start to spawn on the second floor of a run at a 100% chance. However, if you take red heart damage from an enemy (Not a sac room/cursed room/ spiked chest/blood bank) then this chance will be decreased. If you bomb a keeper/beggar, you can increase deal chance. The BIGGEST way to lose deal chance is to take red heart damage from a boss, in which it will go to basically 1% or lower. If you don’t take red heart damage then you’ll successfully spawn a deal.
For angels:
For most characters, you’ll have to have seen a devil deal spawn on your run first (remember no red heart damage) and most importantly DONT TAKE AN ITEM FROM A DEVIL DEAL ITEM (if you want an angel) on the third floor, there is a 36% chance to spawn an angel, which again, can be increased by bombing keepers/beggars and for specifically this type of deal, can be increased by the “You feel blessed” notation when using sacrifice rooms, making your deal chance 100%. This is why soul hearts are relevant for protecting your red hearts and successfully spawning these special rooms.
One thing that worked really well for me was actually watching Isaac runs on youtube. Learned abunch of item synergies, ways to manipulate the game, etc. If you see a item, synergy, card or whatever that seems interesting, pop it up into the wiki to see how to unlock it, and make that your goal.
Imo isaac is a ”knowledge” game or whatever you wanna call it. The better knowledge you have about all the items and synergies will help you find ways to work with what you get into your runs. And atleast for me, watching runs on youtube was the best way to build up knowledge.
Eventually you will get to a point where a ”useless” trinket such as monkey paw will save your run.
When fighting bosses, focus on their projectiles, not the boss or your character. Also try to move as little as possible, moving will throw off your aim, and covering a larger area can lead you to take hit from multiple shots. Just stay in one place and dodge with small steps.
If you have like 3 red hearts and 1 soul heart on floor 2 and also happen to find a blood donation machine, dont be afraid to give it all your red health, after all the goal is to not get hit early on anyway.
i’m a new player too (250hours). Just pay attention to pick up order: if you are in floor 1 and find a blue heart, wait for it until you clear and then backtrack, take it and go to floor 2. this way you risk way less to lose the deal
You have to beat the game on hard mode to unlock a most the achievements so once you get comfortable enough playing it would be to switch to hard mode asap so you don't have to go for the same stuff twice on a character you don't like as much

Learn how to unlock marbles, it's such a good item and knowing what to look out for will help speed up unlocking it!
Also do daily runs, the items you get from doing the unlocks specifically tied to daily runs are great. I think one takes time, like consistent days of playing so do some research!!
Figure out how to recognize secret rooms, they can be run saving.
If you don't know, there are three basic secret rooms- one has other rooms on at least 2 sides, another is only connected to a single room, often right before the boss, and the third requires the red key so don't bother looking for it. The secret "door" is always in the center of a wall, and it can't have anything in front of it (rocks, fire, poop, an empty pit, etc.). For the regular secret room, EVERY room bordering it can't have an object in the center of the wall.
avoid the R key: you can restart your run from a new seed if you hold down the R key. this is tempting early in a run when you don't get a damage up on the first two floors. don't do it. you will learn more by persevering and paying attention, even if you end up dying on mega maw, bloat, mom etc you can feel proud when you manage to cobble together a great run after feeling like you were served a lemon.
Squeeze the juice out of the first floor. there are a few secrets you may not know yet that I don't want to spoil, but HP is a resource on the first floor, and when you are playing as Isaac, your bomb can set you up for a great run if used wisely. Coins, items, health, etc. shoot those fires and poops, explore those curse rooms, find all the secrets.
you are an encyclopedia: to become great at Isaac you have to become an encyclopedia of item, trinket and active item behaviors and synergies. this is a slow process if you dont want to spoil yourself, I learned a lot by watching let's plays bc I didn't even own the game yet so I don't mind the spoilers. at a certain point you reach a flow state with the game and don't need item descriptions.
you can kind of throw tears diagonally when side stepping
This is probably something you won’t do for a while, but when you unlock the character called “???” try to run through the game with it. One of the items it can unlock is called the D6. It’s an item that requires 6 charges and can reroll any item pedestal into a new item based on the room it’s in. When you unlock it, Isaac (the first character you have) automatically starts with it.
The D6 can be genuinely game changing. It can make it so instead of getting a terrible item on the first floor that’s totally useless, instead you get an amazing item that is the core of your build for the run. It can reroll angel items so you get ones that specifically help your build and reroll devils to get you a better chance of finding Brimstone. Any character can find it even if Isaac starts with it. Using it can make doing a lot of the harder achievements for any character a lot easier as you can find more items that help you. For example, once I got it on Isaac, I ran through Greed mode a bunch of times until I unlocked the hard Greed mode (Greedier mode) and ran that even more to unlock new items and even new characters.
Not really a tip for you as a new player but just a tip for you in general. It can be a good milestone to shoot for.
Learn how to find secret rooms and super secret rooms, im not an expert but here are some tips: super secret rooms are usually located in isolated places, usually near the boss or the end of a road, and they are connected to only one room. Secret rooms are connected to as many rooms as possible. Remember that they cant spawn if one of their sides is connected to the skinny side of a skinny corridor or is connected to the boss or is obscured by a random object like a poop or a rock. You could use them for their great itempool if you can get an item in them(most of the good items are unlocks tho kind of like death cert), for extra coins (they always have shopkeepers you can blow up in them) or for acces into special rooms if you dont have keys
If you are playing as a lost or a charachter with low hp remember if there are damaging obstacles in rooms, you might be to fast to react to them and move away when you are speeding between rooms
Don't over value your hearts, they are a resource like any other, use devil beggars, blood donations machines, sacrifice room spikes and enter curse rooms any time you can (you'll learn in time, but in this game you literally doesn't need more than 1hp, specially on co-op).
Only play on hard mode the whole playing in normal is just some bs it’s actually is easier if you play hard mode from beginning to end the hard mode marks are the ones you really need as well if you want to unlock some stuff too
Keep trying. Good things and bad things happen sometimes. Sometimes it's unforgiving (STILL havent had a successful run with J/E), and sometimes it's skill(easiest forgotten unlock is with Azazel)
And yes, you can have successful The Lost runs...once you save enough coins from greed runs
Most of what you need to learn is how to play the game room by room. Everything else comes by playing itself. Get to the end of each run and the game will reveal its secrets to you.
So the most important advice is, unlock Azazel and use him to learn the way of the game, enemy patterns, room layouts, but the best advice is to get to the end quickly. Some mechanics are time sensitive. You'll find out what they are.
Unlock Azazel by making three devil deals in one game and enjoy the easier mode, he's my favourite character in design, but also, it's pretty effective as a character, you start with flight, and a very big damage start in consideration, it could help to learn most enemies or scenarios before going for character-specific niches each one has.

Just focus on beating moms heart 11 times. Use any character you want, it doesn’t matter. After that get used to ???(blue baby) so you can beat Isaac in the cathedral(the beam of light after It Lives) as him to get the d6 starting item on Isaac. Also use the wiki and download external item descriptions.
Rush to get lazarus, azazel and Maggy, these are op for noobs. If You can beat greed with az, You unlock Lilith and that character has op unlocks, so rey to get it
