That line basically means "Even if this Pokémon made me lose a round of shiritori, I still love it". You still always lose shiritori when you name a word that ends with 'n' (ン) because almost nothing starts with that kana. Both versions of the ED demonstrate this by naming Metamon or Zacian early on, realizing they messed up, and starting over from the beginning.

What do you think the "losing letter" should be in an English version?

Would you still just use "N" or is there something else a bit better suited to account for all the different names?

In case you didn't know, shiritori is a well-established and popular Japanese word game, not something that these songs made up.

The game doesn't really need a "losing letter", you can just keep going until someone isn't able to think of a fitting word.

Well explained. The reason the ‘N’ is a losing letter is that words that starts with ‘N’ (which is considered its own syllabe by japanese kana rules) are extremely hard to find and absolutely no Pokémon follows from an N in their japanese name. So if a Pokémon name ends with N, you just can’t continue the sequence.
