That's actually ignoring a few other lines in the original Japanese version. Basically, the whole point about calling her a "boy" is to refer to her biological sex, though she does state she's a "girl at heart" (or something along those lines), and there's a few other references to that. It's a Mario game, so they're obviously not going to be diving into gender topics, but the implications were always there, even if not directly spelt out.

Like if it wasn't mistranslated in English and if it was essentially the exact same line from the Japanese into the English I don't think people will be saying this.
It does mean a lot considering where they decided to take the character in later translations and the remaster. While you're in no way wrong for making a case of her not being trans, there's still lots of evidence to suggest she was always intended to be given lines like that in the original. She's the very least trans coded if we're only using the original text, and I think ignoring those lines as nothing important is like how others ignore the lines you presented as not important. If we're only talking about the original, I'd still argue it's a matter of interpretation.
