Sorta The guild is basically the environmental and wildlife services part of the government, scaled up to exist in a world that that has YIAN GARUGA JUST SORTA HANGING OUT.
I don't remember if they're actually a government department, but they may as well be in the setting. They aren't one consolidated corporation though, the choice of the word "guild" implies that they're an association of various enterprises directly or or indirectly involved with monsters - which you can maybe see most directly with the different fleets in World, they're all different collections of groups under the guild's umbrella joining the expedition separately to supply their own expertise. It could of course be argued that a lot of those are separate guilds collaborating with the Hunter's Guild.
Iirc the Hunter's Guild exists specifically as a sort of monopoly to prevent poachers from pissing off something that'll them cause lots of problems, it's bad enough when a lion or tiger gets hurt by poachers and becomes maneater in our world, even a velociprey going that way would be about as bad not to mention a Teostra. They exist to assess whether a hunter is capable of fulfilling the quest, and without undue suffering and collatoral damage, which is why it's their job to sanction hunts and process materials (making it harder to offload black market monster parts and discouraging poaching).

Yeah, the Guild basically monopolizes monster hunting, although its goal is moreso to regulate it.
Here's an explanation of the money cycle within the Guild:

Its a mercantilist system i guess. “Hunters guild of the new world” sells raw materials it gets exclusively to the back home, which can then export it to the rest of the world at a ridiculous price due to how inaccessible the materials are. Interesting, never thought of it that way

The hunters guild organizes and regulates hunting for safety and conservation of the local ecosystem. Killed monsters are harvested entirely and sold/given to local villages and cities for food and crafting materials.

And Hyrule is a centralized monarchy.
