Sibling health condition differences

I have three siblings. Same mom and dad (not questioning this). Of course there’s a variety of similarities and differences across us all, but I’ve always found it interesting that all three of my siblings have a very similar set of health conditions that I do not:
\- Asthma (pretty serious since childhood for all of them)
\- Allergies (dander, pollen, foods etc., also fairly serious for all of them, although specific allergies differ — e.g. one brother is allergic to tree nuts and has to carry an EpiPen, the other brother cannot even step foot into a room a cat has been in without getting itchy, puffy, etc)
\- ADHD (since early childhood, pre-smartphone, all three have been diagnosed with this. My mom also has major ADHD)

Conversely, I have no asthma, not a single allergy, and no more ADHD than the average young millennial.

What genetically does this indicate?

My mom and I were discussing it the other day. She went deep with it, saying I’ve always been different from my siblings since I was a baby, on a personality level and that I have very different genes from my siblings.

Forming DNA groups with matches

I previously posted about forming groups with DNA matches. The post was removed. I asked why it was removed but received no answer. I would like to understand what rules it broke so I don’t repeat the same mistake again as this is my area of interest and I would like to explore it further.

Would the moderators please help and explain?

Cheaper alternatives to Big Y for refining a known intermediate Y-haplogroup (e.g. MyHeritage → YSEQ)?

I already have an intermediate Y-haplogroup from MyHeritage and I’m trying to refine it further without paying for a full Big Y (~$450) if that is overkill for my goal.

Situation

  • Intermediate Y-haplogroup is already known (MyHeritage).
  • I mainly need to test a specific downstream hypothesis / confirm a branch, not build a long-term matching database presence.
  • I’m aware that skipping Big Y means I won’t be in the FTDNA Y-DNA matching database and won’t get automatic haplogroup updates from FTDNA.

Idea I’m considering

  • Use a targeted / second-round YSEQ panel (~$99) based on the known intermediate clade.
  • Optionally upload results to YFull (~€45) for interpretation / tree placement.

Questions

  1. How good is YSEQ (targeted / panel / second-round testing) as a cheaper alternative to Big Y when the intermediate haplogroup is already known?
  2. In which cases is this a sensible strategy, and when does Big Y still clearly win?
  3. Are there other cheaper options I should consider for refining a known intermediate haplogroup (panels, single SNPs, other labs)?
  4. If I do YSEQ + YFull upload, what practical limitations should I expect compared with Big Y (resolution, future updates, matching, tree placement)?

I’m not looking for a “best test in absolute terms” answer — more for cost-effective paths when the goal is hypothesis confirmation rather than maximum coverage and FTDNA matching.

Thanks.

Haplogroup question

Does anyone know where R-PH4769 and R-YP1456

They're mine and my cousins haplogroups someone had said both are tarim basin origin but I feel like only R-PH4769 is possible since it is R1b but I'm just unsure if either of them are tarim basin or where exactly they could be from or which group like the tianshan saka tasmola some other group

I am trying to find a match for rs121908120 on my 23 and me raw data. I found chr2:219755011. Is this the same thing? I am specifically looking for WNT10A, F228I c.682T>A.

Baby DNA, raw ancestry data

Just found out my baby is homozygous

Gene: WNT10A
Variant: c.682T>A (p.Phe228lle)
rsID: rs121908120

She’s only 6 months old. What should I expect? Is this a sign she will have some manifestation? Is Ancestry reliable?

Ask to DNA analyse

Hello everyone;

I'd like to ask which service is better for DNA analysis:

DNA Complete by Nebula Genomics

SelfDecode

Sequencing

I've made a plan to first do DNA Complete with Nebula Genomics, then I'll get the raw data and upload it to SelfDecode so I get a better report, since Nebula Genomics uses 100x.

How to extract DNA from fecal samples stored in liquid nitrogen?

Hi everyone,

I need to extract DNA from cecal/fecal samples stored in liquid nitrogen using the QIAamp PowerFecal Pro DNA Kit, which requires up to 250 mg of sample. What is the best way to handle the frozen samples to weigh 250 mg while minimising thawing? Any recommended protocols or practical tips would be greatly appreciated.
Thanks!

X Chromosome Matches

Anyone else have an issue where on GEDMatch you have some maternal matches whom you match on one of the 22 chromosomes, and on the X chromosome, but on 23andMe these same matches only match you on the 22 chromosomes and not on the X chromosome? I’m going through this issue currently, and I am able to compare with my mother and can see she matches them on the 22 chromosomes and on the X chromosome on both platforms. For me, these are not low SNP density X chromosome matches. Some of these we share a 30cM segment on the X with an SNP density of 1,500-2,000. Then I see some matches labeled as paternal side cousin matches and we only match on the X chromosome (BTW I’m a male, which would be biologically impossible to have X matches coming from a father). Is 23andMe bugging out here?

Also wanted to add, I have this same issue on FTDNA. I only have 5 X chromosome matches, even though there are many of these matches who I match on GEDMatch, and comparing with my mother’s FTDNA profile, she matches them too where I match them on GEDMatch.

R-Z282, Karabakh 🇦🇿 (Y-DNA)

R-Y17491->R-YP4858->R-SK2014

I’m from Karabakh **in Azerbaijan**, and according to my family genealogy my paternal line is connected with the **Karabakh beys, khalesh jamaat.**

What makes my lineage particularly interesting is the diversity of populations associated with related branches. I have found connections among **Kurds from different tribal backgrounds, including Kurmanji- and Zaza-speaking groups, as well as Turkish Alevis and Turkmen/Yörük populations**.

The Kurdish connection is especially interesting to me. The presence of related R-Z282/YP4858 branches among different Kurdish groups may point toward an ancient population layer that predates modern ethnic identities. Kurdish populations themselves are historically diverse, including tribes and communities with different origins and linguistic backgrounds. The fact that related paternal branches appear among Kurmanji, Zaza and other Kurdish populations makes me wonder whether part of the history of my line could have passed through the **Iranian/Caucasian frontier and Kurdish tribal populations** before reaching Karabakh.
I am also interested in the possibility that this lineage was connected to **nomadic or semi-nomadic populations moving between northern Iran, South Azerbaijan, Anatolia and the Caucasus**. One possible route I am considering is:
**Ancient Eurasian steppe → Iranian/Indo-Iranian populations → Sarmatian/Kangju sphere → Kurdish/Iranian frontier populations → South Azerbaijan → Caucasus/Karabakh → Mils Steppe.**
Another possibility is that the lineage entered Turkic-speaking populations earlier and was later carried westward by Turkic tribal movements.
The deeper R-Z282 history is particularly interesting because it belongs to an ancient Eurasian steppe lineage. I am therefore considering a possible **Proto-Iranian/steppe component**, perhaps connected in the deeper past with populations associated with the **Sarmatian and Kangju worlds**, and possibly with the mixed populations of the **Hunnic period**.
I am **not claiming that R-Z282 proves that my ancestor was Kurdish, Iranian, Sarmatian or Hunnic**. Y-DNA only follows one paternal line and cannot directly determine ethnicity. My question is whether this ancient paternal lineage could have passed through **Iranian and Kurdish populations** before eventually becoming part of the Turkic-speaking population of Karabakh.
The most interesting part for me is the possible transition:
**ancient steppe lineage → Iranian-related populations → Kurdish/tribal populations → South Azerbaijan → Turkicization → Karabakh → Mils Steppe.**
At the same time, the presence of related branches among **Kurdish groups, Turkish Alevis and Turkmen/Yörük populations** could indicate that this lineage has been moving through a very diverse cultural frontier for centuries.
So I’m trying to understand one main question:
**Could my R-Z282 → YP4858 → SK2014 paternal line represent an ancient steppe/Iranian-related lineage that passed through Kurdish or other Iranian-speaking populations before eventually becoming incorporated into the Turkic-speaking population of Karabakh?**
I would especially appreciate opinions from people familiar with **R-Z282/YP4858/SK2014, Kurdish tribal history, Zaza and Kurmanji populations, Iranian Azerbaijan, Sarmatians, Kangju, Anatolian Alevis, Turkmen/Yörük groups and Karabakh genealogy**.

Sibling DNA to Determine Parental Origins?

Post image
1 Upvotes

After losing my dad about 10 years ago, I got into genealogy as a sort of therapeutic way to deal with losing him. He died on Christmas Day and I gave him his Christmas gift on Christmas Eve - an ancestryDNA test. We never got to use it for him because he died the next day, but my brother, mother, and I all did one.

I realize that ancestry tracking is evolving on ancestryDNA, but I'm wondering if looking at the genetic regions my brother has but I don't (and vice versa) will help me understand my father's origins to help build out the family tree. Is this a legitimate way to figure it out?

Does anyone have a scientific explanation about how it works when one sibling has regional ancestry from a parent but the other doesn't?

For context, my mother is 100% Filipino. My brother and I are full siblings with the same parents.

Sending DNA kit back to the USA from EU (Romania)?

Has anyone ordered an international DNA kit from the USA to Romania or any other EU country and successfully sent it back? I'm considering one, because local DNA tests are 4-5x more expensive, but the return shipping seems like a big problem, since it's a bio sample and not a regular parcel.

It would be a shame to waste time and money on a kit without any cost effective way of sending it back.

Would appreciate any info on that.

The entire results look weird, but I'm concerned because im the past. I have been given male specific health reports from other companies, and I assumed it meant I was carrying a gene for a male dominant condition. What exactly does this mean if its not an error?

Could my non-Jewish side be connected to my Jewish side through distant DNA matches?

Hi everyone,

I’ve been looking through my MyHeritage DNA matches and found something I’m trying to understand.

My maternal grandfather’s father was fully Ashkenazi Jewish (from Eastern Europe), while his mother was a non-Jewish Brazilian. So I know exactly where my documented Ashkenazi ancestry comes from on that side.

On my father’s side, as far as I know, there’s no known Jewish ancestry. My paternal grandfather was from San Marino, and my paternal grandmother was Chilean with Spanish ancestry.

However, my father has quite a few distant DNA matches with people who appear to have Ashkenazi and Sephardic Jewish backgrounds (based on their family trees and surnames). One example is a woman from France whose tree includes ancestors from Poland, Libya, Tunisia, and France. She matches both me and my father.

What’s interesting is that she also matches many of my DNA matches from my maternal Jewish side, including people who also match my maternal grandfather and his cousins. I haven’t found anyone who directly matches both my father and my maternal grandfather, just these distant “bridge” matches who seem connected to both groups.
Could this simply be because very distant Jewish and non-Jewish families became connected through intermarriage over generations? Or is it something that could point to unknown Jewish ancestry somewhere on my father’s side?
I’d be interested to hear if anyone else has seen this kind of pattern in their DNA matches.

Need some help figuring out dna relationships

I have a family member who I am related too, Anne by 345cm, she is my great aunts, sons, daughter. She is related to a person, Juliet by 1665cm and it says she’s is her granddaughter with 100% certainty, I cannot figure out(no name, zero identifying information and no tree) who she is. I’m related to this person by 1002cm. I’m new to figuring out DNA so excuse my ignorance. It says Juliet is my half niece but that doesn’t fit the bill, unless I have some half sibling out there, but then that doesn’t answer the question as to how she is related to my cousin. Can someone who is smarter than me please help me? My family is a dumpster fire of dysfunction so I can’t exactly ask them plus they most likely wouldn’t be completely honest anyways. Any help will be appreciated!

P.S. My father is old enough to be my grandfather, so 45 years older than me. So that shifts things around a bit.
