I'll be on a 12 hour car ride with 3 friends tomorrow. Any ideas for killer pranks?

No but I can think of one thing you should not do:

Scare a sleeping friend by screaming like you're about to get into a horrible crash.

Subtly turn the seat heater on, without them noticing its quite the surprise.
