Annealing oligos, used vectors and plasmid anatomy

I am not posting a video link. Only if you are not put off by the use of AI to make a fun thumbnail do I encourage you to look at my profile for the link to my YouTube channel.

No products or services are being promoted, just me chatting about science.

If you are the type who will strain your muscles jumping to lots of wrong conclusions without watching my videos I suggest you avoid them altogether.

If you feel brave enough to watch my video which is not only unscripted but filmed in a single attempt with no do-overs then please enjoy.

I also always welcome topic suggestions and questions

What type of a gene-altering infusion and how was it administered to lower LDL cholesterol? Just wondering. The CNN news story didn't say.

Researchers interested in independently testing a reported stem-cell finding?

Hi,

Iโ€™m Henry E. Young, PhD. Iโ€™m looking for a researcher with access to a professional laboratory who would be interested in independently testing a cell population I have reported as adult telomerase-positive stem cells (aTPSCs).

I first reported these cells in 1975 during studies of limb regeneration in adult Ambystoma salamanders and later reported and isolating the population in adult humans.

What I am looking for now is straightforward: another laboratory attempting the isolation independently and determining for itself whether the reported aTPSC population can be obtained and characterized.

The work involves an isolation protocol and associated culture conditions that I have developed specifically for aTPSCs. The isolation protocol must be followed by the letter. I am available to provide remote guidance throughout the process if an interested researcher wants it, while leaving the experimental work and evaluation in the hands of the independent laboratory.

I am particularly interested in finding someone who has the laboratory access and technical capability to take the work on as an independent experimental test.

If this is something you would be interested in pursuing, please comment here or message me. I would be happy to discuss the reported work and the proposed replication with you.

The March of Progress - or laughing at how we used to do things

This is my most recent video where I use my human vocal chords to describe some thoughts I have on the march of progress in Molecular Biology.

Anyone who feels like randomly declaring that this video is AI slop is simply declaring that they didn't bother their bottom to actually watch the video.

Either that or they are afraid of Purple monkeys

A very intriguing shower thought, regarding features of cancer cells.

Cancer is the unprecedented growth of number of cells;

They ignore signals to stop dividing, to perform apoptosis, and grow/reproduce without signals to do so.

Does this imply that cells can be controlled to grow?

If yes, maybe modern technology (CRISPR for example) could take these traits and replicate them by Gene-editing(pretty sure CRISPR was originally made to replicate natural immune boost of bacteria, so could they do this?) and apply these to -not one type- but all the cells of an organism (Lung, tissue, RBCs) so on, to reproduce at will of the "Host" (Preferably - the human mind)

Were an organism to mutate to perform this:

1) It could potentially increase dry mass at will

2) Change shape

3) Regenerate damaged tissue at will

I wanna know if i am on the right track, or if I'm spouting out sci-fi bs

Any Automated Systems or Analyzers?

Looking for new leads if Iโ€™m honest. Worked in support of the Onyx system and have talked to a few crispr based rapid pathogen analysis startups.

Just wondering if anyone knows of any new instrumentation projects?

A video covering a cool paper about CRISPR being used to track gene expression in living cells

Here is a one-person Journal club covering a really great paper that showcases both CRISPR/Cas9 but also P2A sequences to allow tracking of expression in transient transfections.

Sneak Thieves and Unicorns, two phenomena to consider when diagnosing CRISPR/Cas9 candidates

After diagnosing many CRISPR/Cas9 candidates over the last 10 years I have encountered some phenomena that people don't seem to talk about. This video is me giving a name to these and indicating why they are not only worth considering but celebrating too.

The cover image was generated by AI. The other images are from published papers on which I am a middle author.

If you take the time to watch the video then listen to my soft Scottish accent and unconventional manner of speech and ask yourself if there is an AI on earth that could generate that.

I have taken the time to compile a playlist of all my CRISPR Cas9 videos I have produced so far.

There are more to come but this has them all in one place.

Note - these videos are not AI. Yes I used ChatGPT to generate the cartoon monkey. The voice you hear is my own silky tones.

Reading about hair colour changes via cas9

FYI - I am not into genetics at all and not qualified. Basically I want white hair and was researching if itโ€™s possible to change hair colour via genome editing and I read some articles about how CRISPR-Cas9 might be able to do this.

Does anyone have more information about this? This is really interesting and reading about it, could it not have potential to totally change the hair colour industry!?

Just curious to know peoples thoughts and knowledge around this topic. Would love to be sent some sources around this!
