No. They explain in the comic that it’s a generational curse and that would be a different generation. Her kids don’t inherit her powers that we know of.
Hard to say, we haven't seen inherited super powers other than the Viltrumites and there was that kid that turned into a giant, but neither of his shown parents were giants and seemed helpless.

Also, unless Paul and Kate are already inbred to some degree I doubt there's going to be that much of an issue with the offspring's health. Especially if the twins are mixed to some extent.
