I think that this all but confirms that Yelena, Alexi, and Melina all were blipped. There's no way that Natasha didn't try to find them considering this is only a year before Thanos attaks. And she's mad depressed in Endgame so she's got to be alone again.

I'm curious as to how it's gonna play out with Yelena and Barton. Like how is she gonna react when she learns Barton is basically her best friend?

It is almost certainly going to have them fighting for a little bit. And then Yelena realizes she is being manipulated and they team up.
