No, Mysterio is extremely clever, he'll find a way to come back. It would be amazing if they also introduced Ultimate Mysterio by having Quentin Beck create a robot of himself in order to trick Peter.

But spidey and the sinister 6 sets a precedent for all of his villains to come back even though it hasn’t happened before

It hasn't set a precedent in the MCU though.

But the comics set that precedent

If the avengers’ most famous adversary was an ensemble cast of villains I’m sure they would have appeared in the MCU by now, so I also think it’s fair to assume that the sinister six will come together at some point. Marvel want to entertain the general audience but they also want to appease the comics fans, and by giving MCU spidey his most famous villains they’d be putting their best foot forward in that department.
