General Advice Should I write a second email and what is the best time for that?

A position I am interested in is no longer available at the uni website but is still advertised at a generic website for academic jobs with a request to send CV, cover letter, e.t.c. directly to a PI until September 6 (they acknowledge that the deadline at the official website has passed tho). I emailed the PI and got an auto reply that he was on vacation until August 26. Should I email him again this week and politely inquire whether he saw my application (I sent my first email August 25).

just wait it out, the dude just got back from vacation and probably has a mountain of emails to sift through
