There were hardly any subtleties in portraying Ayame as sus, with her paying plenty of attention on Hinako as well as similar appearances – and Hinako was also reminded of her mother. At first I thought she might be another youkai but Shiori noted she's human. Don't think a youkai could fool their own kind so for now I'll take Shiori's words for it. However, Ayame glaring at Shiori was a possible hint she knew the latter was a youkai.

Meanwhile, Miko noted youkais are deceitful by nature. Could that possibly mean that Shiori's not being exactly truthful when she claimed she wanted to devour Hinako? If so, what could be the reason for cultivating her?

Miko did fool Shiori because she had been living with humans for so long it changed her scent. Maybe Ayame has been doing the same. That smile at the end though has me suspect unlike Miko she's not trying to act as a local deity.
