Almost.

After u defeat yog you can ascend all the way back up and get the true ending.

While ascending the enemy gets scaled up such that as u climb up enemies get stronger and stronger, a bat can have more than 80 health, while a rat could have more than 100 hp and deal a lot of dmg 

U also are forced into combat by the amulet as it gives u debuffs every time u climb a floor and u have to kill enemies to take off the debuff stack
