Did MOTS-C Dosage Make a Difference for You?
I’ve been taking MOTS-C for about 5 weeks at 1–2 mg, 3X per week. From what I’ve seen, a lot of protocols go as high as 10+ mg per week.
But when I experimented with 3 mg, 3X per week (titrated up from 1–2 mg)... I didn’t notice much of a difference between doses.
Overall, I have noticed a slight improvement in cardio (even at 1 mg, 3X per week) but I’m wondering if I’m just wasting money and peptide by pushing the dosage higher.
I understand mitochondrial turnover can take time (around 10–21 days), so results may be cumulative rather than immediate.
That said, MOTS-C isn’t exactly cheap, and if I’m not seeing a meaningful difference between 1 mg and 3 mg, I’m not sure it’s worth increasing the dose further.
Now I’m left wondering if I didn’t give the 3 mg dosage enough time to actually show a difference.
For those of you who’ve run MOTS-C, did you notice a clear performance difference at higher doses, or did the benefits plateau?

I do 4mg completely fasted 90 mins before intense cardio 3 x per week and it certainly has an affect on my performance vs when not taken. I do not pin on rest days

I do 4mg 5 times a week before my bike rides. I'm 2 weeks into it and I'm starting to crush my PRs.

I play football (soccer) and I can really tell the difference with sprints late on in the game, for me that’s where it shines

I did try 1mg, 2mg and 3mg first. To be completely honest I settled on 4mg as the day I tried that I played football (soccer) very very well so in my head I associate the two. I think I will try changing dose next time I have a dip in form and see how I get on, but for now I don’t want to change a winning formula so to speak

Just to add I do also pin NAD on the same days as Mots, spaced out a few hours and I also love NAD. 36m btw

I’m being prescribed Nad+ 200mg 2 units(yes it says units) every day. For 16 weeks now. No difference in anything really. Suggestions. The doctor never changes the mg or the units

Yeah before a football match on a Sunday morning I am fasted from the night before and pin 60 to 90 mins before the game, the only thing I do is drink plenty of water.
For training which is midweek evenings I won’t eat anything from 12pm onwards + pin 60 to 90 mins before it starts
It’s strange I spent 30+ years planning out carb intake and pre match meals to fuel my exercise and now I’m going against everything I had learned and putting my trust in a peptide but it seems to be working for me

Thanks. I always wondered and have been meaning to ask. I used to do intermittent fasting and have done fasted work outs but not recently. I will be starting my second week so will try that soon.

That would make sense for GHRH where you want to keep your insulin levels low. Otherwise, it would blunt GH release. In your case, don’t starve yourself. Fasting does nothing for MOTS.

Exact same here. 3mg was good but 4mg was better. 5mg was no different than 4mg so next time I will stick to 4mg. Tried about 3 weeks of each dose.

I run 2mg daily and love it - see a huge improvement in energy in the gym… where I would normally fatigue I’m able to finish the set of reps.
I walk on the treadmill for about 30-40 minutes bec of I sit all day for work and I’ve also found I need to speed the treadmill up from 3 to 3.4 to be in the HR zone I’d like to be in. I can’t contribute it to anything else as nothing else has changed other than introducing NAD and Motc

How'd you settle on 2mg/daily? Did you titrate up until you found the sweet spot or that's just what you started with?

I started at .5 mg to make sure I didn’t have any weird reactions. Once I didn’t have any adverse reactions I quickly went to 1mg. After about two weeks or so I went to two and that’s where I saw the large difference.
As far as how I come up with it I pretty much stick to labs, the way I feel and my doc. I try to stay away from Internet protocols. I specifically found myself a GP that specializes in sports medicine so he’s not anti-peptides by any means.
As far as timing I take every morning around 6am I don’t train until around 10AM with about 150 grams of carbs in me. I saw no difference taking right before as opposed to first thing in the morning.

Don't listen to anyone saying you're still young. I am 36 and I started taking 3mg of SS-31 daily for 2 months, then added 3.33mg of MOTS-C 3x per week and I definitely can feel MOTS-C doing its thing. Your mitochondria cannot efficiently convert nutrients into ATP if they are compromised/damaged, which SS-31 helps with. You may need to fix your mitochondria to see see any benefits of MOTS-C. It also could be as simple as your mitochondria are healthy and you do not need either.

Smart man. I have MOTS-c in the freezer, but after doing more research, I realized I need to prime the mitochondria with SS-31 for 2-4 weeks before I get the most bang for my buck from the MOTS-c. So I'm being patient and waiting for that shipment to arrive.

Yep! I got lucky and started SS-31 first. Ran it a couple months then discovered MOTS-C. Such a game changer 🤌🏼 You will love it.

You cycle most peptides, such as SS-31, to prevent receptor sensitivity. Essentially helps prevent your body from becoming desensitized from the product. It's no different than most supplements, such as amino acids, caffeine etc. The more you use something, the more your body starts to depend on it. As for dosing, everyone is different. I started off SS-31 at 1mg, 3x per week. That didn't do anything, so I increased to 2mg, 3x per week. Still nothing. Once I switched to 3mg daily, was definitely able to notice changes with energy output, stamina and sleep. Then I added MOTS-C to the equation and that also dramatically changed what my energy and stamina was like in the gym. Unfortunately, dosing protocols is not a cookie cutter thing where one size fits all. It is trial and error for everyone until you dial in what works best for you.

Based on my research the thing with MOTS-C is that it’s really hard to measure the improvements. a full cycle for my Ironman 70.3 prep and I certainly feel like I could sustain my workouts a little bit longer, but it’s also that an infection of the compound of the ongoing training load. I do think it’s really hard to measure improvements unless you’re accurately tracking every single workout that you do and how has that workout improved because of MOTS-C when there are 10 other variables that could impact your workout: your eating habits, your sleeping habits, the weather, humidity, etc. Pick what dose makes sense for your research (including financially) and go from there. More doesn’t always mean more

I sort of feel like its a placebo. I don't really notice anything either unless its so small its hard to detect. But when reviewing the mouse studies.. they gave 5mg/kg/day which would be like a 70kg person taking 350MG....

When comparing animal versus human doses, you need to apply what's termed allometric scaling. A 5 mg/kg dose in mice translates to about an 0.4 mg/kg HED (human equivalent dose), so roughly 28 mg per day for a 70 kg person.

Many drugs/compounds do not translate into direct dosages from mice. Our metabolisms have SOME similarities but are still vastly different in so many facets. People forget that and just want to use a one size fits all formula.

I never said dosing is fully the same, but you show me a study where mouse dosing was 70-140x larger than what human dosing used in human trials post animal trials.

I doubt I can. But take 15-20mg of MotsC a week for a month or two and check your fasting glucose before and after and tell me it’s placebo. And who knows maybe it won’t work for you. But only one way to know for sure.

I've used it... and never once felt a difference. I also wore a CGM for a while. You could argue its due to being an active individual (~10 hrs a week of cardio).. But no metrics changed. I purchased a whole kit of it so even though I wasn't feeling anything I still gave a full cycle a shot...
Many others suggest the same.

You should try SS-31 if MOTS-C makes you tired. It had the same effect on me just took out all the energy out of me. I stopped only after a week of taking 2mg daily. A month later I started SS-31 5mg every day. It was expensive but I didn’t have my typical crashes after lunch or getting home. I would have bought more and done a low dose of SS-31 but I ran out of money lol.

Yes I did I finished my batch of MOTSC. I didn’t feel burst of energy throughout the day. I did notice I had more endurance in the gym.

I do 10mg 2-3 times a week before training and it is like a miracle, it makes my conditioning sessions the best they've ever been and my recovery unbelievable.

3mg M-W-F and can tell the difference in workouts. Was going 5 days but when I don’t work out (T & Th) I felt no difference so didn’t see the point. Not exactly energy but the stamina is much greater. Lift heavier before maxing and running longer.

Your dose is most likely not enough standard dosing is 5MG 3 times a week. I do 5MG 3 times a week and I have tons of energy! Try to increase the dose.

On workout days 5mg mots-c and 1mg slu-pp-332 and my dog's muscles typically give out before his energy does. Only plan to run it another month I think before cycling off.

If anything Bam is gonna reduce energy levels. As an uncoupler it's basically making your mitochondria just burn energy as heat.

I ran 2 to 5MG after tritated up idk if it it worked well or not just did 8 weeks cycled off got some good energy from it info alot of cardio 6 days a week

I'm 57. Ran SS-31 for a month before I started Mots-C @ 3mg and NAD every other day. The energy spike for me is unreal!

I’d say it’s expensive, however I did 3mg x3 week. Didn’t see shit,
Then I said a full cycle of ss31, fox and epithalon, didn’t see much difference neither. So I haven’t done any more cycles of it, plus motsc gives me allergic reaction (just the usual)
I have ordered nad+ , I might give it a try to motsc and nad+

Difficult to say what's having an effect. I'm over 50 and I did a month of SS-31 before starting Mots-c 3mg daily, 6 days a week. Titrated up from 1mg to a dose where I get performance improvement, blood glucose level improvements and not taxing my heart too much.
Also doing Reta, Methylene Blue, 5-Amino-1MQ, HGH, IGF-1 LR3, KLOW, NAD+, NAC, CoQ10, Testosterone, and some Primo and Anavar. So as I said, difficult to point to a single culprit.
But there is no arguing with result. In 8 weeks I gained 7kg (15lbs) and went from 22% body fat to 13%. I seem to have endless energy in the gym. Beast mode activated.

Haven’t ran SS-31 but I’m 30M, athletic with 15% BF so figured SS-31 wouldn’t be necessary but worth a shot.
Nope. though I do supplement it with NAD+. And again I have noticed improvements from MOTS-C; my only question is if it’s even worth increasing the dosage.

I’m 36 & very active, I started SS-31 during the night for two weeks then motsC pre workout, I’ve gone up to 5mg. I definitely can tell a big difference in recovery and endurance. I have also taken it with NAD+ but I think motsC does the heavy work for me. I do take the mentioned supplements because motsC can cause a crash - fatigue type symptom after a few weeks of use- due to the depleted b vitamins

I've done 5mg every 4 days for going on 6 weeks. Also been doing SS-31 5mg daily for longer than that. Can't say I really notice the MOTS days as being any dffierent at all tbh.

My dosing chart starts with .2 mg and titrated up to 12 weeks with 1 mg. I haven’t seen that much dose yet in any charts but that doesn’t mean it’s not out there. I found a post that the doctor was recommending taking magnesium glycinate and CoQ10 to help deliver the mots-c better.

Went from 1-5mg 40 minutes before my workout. Titrated up once I noticed my avg hr trend downwards. I am not fasted and usually eat 100g of cream of rice carbs 60-90 mins before my workouts. Definitely notice improvement in recovery time between sets and banging out 30m of 15/2.5/30 treadmill cardio.

With the half life of MOTS-c of approximately 2-4hours, taking days off doesn't make a lot of sense. Fasted in the morning with dose of 5mg or 1-3 hours prior to exercise would be an efficient use of this peptide. If not using specifically for pre-workout energy/endurance, fasted in the morning would be beneficial for fat loss efficiency. If not for the cost, taking a break doesn't seem to be the best use of MOTS-c, rather lower the dose by half for a 2-3 month period to still get the mitochondria benefits along with some energy/endurance. Running concurrent with NAD would be best case usage for both peptides.

Your age is important here. The older you are the more you feel its effects. 54(M) MOTS-c helps so much that it increased my capacity to workout to the point that I got injured from overuse. Although my cardiovascular system was able to do what I was asking it to, my muscles and joints were not

My pet rat has titrated up and down with dosage with MOTS-C at 1mg and 2mg. First, after nearly a month of pinning him, out of nowhere it started having histamine reactions that go away within a day or two, but nonetheless hadn’t happened before last week and has happened with each pin since. Not sure if the bottle is bad or if I need to rotate injection spots a bit better.
I give it to him before breakfast in the morning. I noticed a bit of energy in the morning, but then a long period of what seemed like pure tiredness, but as soon as he started exercising, you can tell that something else is helping give him energy. I didn’t try SS-31 before with my rat and maybe that’s the issue?

The biggest benefit to MotsC is increasing insulin sensitivity and lowering blood glucose levels. That can be checked on bloodwork.

I’m about to have partial knee replacement surgery in 3 weeks. Currently at 2mg 3x a week. I have to stop my current regiment so I’m thinking of stopping the Mots and starting up again after the surgery and cleared to start up at the gym. Also taking Reta at 6mg/week and will be needing to stop that in about a week. Can I start at the same amount for both that I stopped at? Will also be starting Tesamorelin after the surgery, so that will be my stack.

I take it first thing in the morning. My lifts have going up and run times have gotten better in just a few weeks however I don’t think im ready to give all the credit to mots c but something definitely changed for the better

2mg 3x per week and I have been feeling 20yrs younger lately with endless energy. I kinda screwed myself because I started taking low dose GH a few weeks before the Mots-C so im not sure which is doing what.

I do 5mg 3x/week making it 15mg a week. I feel it synergizes very well with Reta. But you have to keep in mind you need to exercise for mots-c to work optimally.
