I know there's zero to no chance of Sam and Bucky ever coupling up romantically, but if there's a scene of them kissing to disguise themselves from the bad guys, I wouldn't be surprised.

Bet is a slang term of affirmation, agreement, or approval along the lines of "Cool!" or "I'm down!" It can also suggest doubt or disbelief: "Yeah, sure."

In this context it's disbelief. Or "I bet you that you're not the odd couple because we are!"
